What, specifically, are you looking to get out of the device and which device are you considering (the basic e-readers or the tablets?)
I have a Nook Simple Touch, and I love it. Though it's main interface connects you to the B&N store, I like that it has epub and pdf support that allows me to access ebooks from a variety of outlets (including public domain titles through Google Books). The design of the device is excellent. It's very lightweight and comfortable to use for long periods of time, and I appreciate the long battery life.

I have the last generation Kindle, and I really like it. I got it because my campus was cutting back on printing and I could read all my students papers and all my research pdfs on it. It looks like a book page when you read off of it which is nice, it helps cut back on my staring-at-screen hours. The only thing I didn't like about it was that it wasn't a touch screen. I guess the newest Kindles are.
Honestly most E-readers are the same.

I adore my Color Nook. I hate the flash/refresh of eink readers. I use my Color Nook far more than I ever imagined I would. I am borrowing books from the Free Library of Philadelphia on a regular basis and downloading the Free on Fridays books from Barnes and Noble (no nook purchase required). My Color Nook lets me read a ton, surf the web, and check email. Geek bliss.
One thing i like about the Nook rather than the Kindle is the format of the files. Amazon is trying to lock things down. The Color Nook uses the epub format which is more universal.
This, from a VERY reluctant ereader adopter.

I have the old black and white nook. I love it. I use it only to read, so the color doesn't really appeal to me. I got it before the Kindle could be used with the library books, which was one reason I went with the nook.
I don't know the differences now. I do recommend an ereader, though. You will love it!
